The research opportunity
Barrettin is a marine natural product first reported in 1986 from the sponge Geodia barretti. Structurally, barettin contains an unsaturated brominated tryptophan residue and an arginine moiety cyclized into a 2,5-diketopiperazine (DKP) core. DKPs are known to exhibit desirable physicochemical profiles and serve as privileged scaffolds for medicinal chemistry optimization and barettin presents a functionalised core for late-stage diversification. Cartmell in 2026 reported barettin to be an inverse agonist of 5HT2a with antihyperalgesic properties and represents promising lead in the treatment of chronic pain.1 The research project aims to combine biocatalytic technology and medicinal chemistry to develop a series of barrettin-like analogues for 5HT2a inverse agonist activity. This will utilise our recent discovery of a flavin dependent cyclodipeptide oxidase from the halophile Nocardiopsis dassonvillei (NdasCDO) to oxidise diketopiperazine motifs2 selectively to enable rapid access to barettin-like scaffolds.
Research approach
The first phase of the project will be to develop this key biocatalytic step to demonstrate applicability of this approach and optimise conditions for both substrate generality and throughput. Once established a library of barrettin-like analogues will be synthesized to probe structure activity relationship studies to probe structural features necessary for 5HT2a activity.
Supervision and collaboration
This is a multidisciplinary project involving biocatalytic reaction development and medicinal chemistry under the supervision of Dr Gordon Florence and Dr Clarrissa Czekster at St Andrews and in collaboration Prof Christopher Cartmell at the University of Arizona.

PhD: A Biocatalytic Approach to Barettin-Inspired 5-HT2A Inverse Agonists
Quick Decision
Promising with material uncertainties. This St Andrews Chemistry project joins a defined medicinal-chemistry question to a biocatalytic platform recently studied by both named St Andrews supervisors. EastBio adds cohort training and a professional placement. The advertised work is broadly clear: assess the relevance of the enzyme-based route, then investigate Barettin-inspired compounds and their relationship to 5HT2A activity. The most important unresolved condition is financial: EastBio describes home-level tuition support, while St Andrews publishes a 2027 overseas Chemistry research fee of £28,140 and the project does not explain any international fee top-up. EastBio also caps overseas awards at 30%. The deadline is 16 November 2026, only ten calendar days after this 6 November review date, and late applications are not accepted. Contact the project supervisor and St Andrews administrator immediately about fee liability, eligibility and submission details.
Research Problem and Doctoral Work
Barettin is a marine natural product whose structure contains a cyclic dipeptide core. A 2026 primary paper reports Barettin as an inverse agonist at the 5HT2A receptor and describes antihyperalgesic effects in a mouse model of chemotherapy-induced peripheral neuropathy. The study reported effects in male mice but not female mice. This is a preclinical rationale for further work, not evidence of human efficacy or a treatment outcome. PubMed’s record of the 2026 Barettin study
The project proposes to combine biocatalysis and medicinal chemistry to study a series of Barettin-inspired analogues. Its broad research sequence is to assess whether a recently studied cyclodipeptide oxidase can support access to the relevant compound class, followed by analogue research and structure–activity analysis around 5HT2A activity. The source description names substrate scope and throughput as questions for the first phase, but it does not specify how many analogues, the assay portfolio, milestones, decision criteria or the division of work between partners. Those details should be discussed with the supervisors rather than inferred.
The closest scientific support is a 2025 Nature Communications paper coauthored by Gordon Florence and Clarissa Melo Czekster. It characterizes NdasCDO, the enzyme platform named by the project, and reports broad substrate scope across cyclic dipeptides. This makes the proposed research tool credible and gives the student a direct link to recent team work. It does not establish that the project’s particular analogues will be accessible or biologically active. Sutherland et al., 2025
The scientific value therefore depends on connecting the chemistry to a clear pharmacology plan. A useful interview discussion would establish which partner conducts biological evaluation, whether the student will take part in that work, and how the project will interpret negative or mixed receptor-activity results. The 2026 mouse result also makes sex-dependent evidence and the boundary between receptor activity and analgesic effect relevant questions. No therapeutic or clinical claim should be expected from a chemistry PhD.
School, Programme and Field Context
The University of St Andrews School of Chemistry is the research host and offers a full-time Chemistry PhD normally lasting three to four years, with a thesis and oral examination. The supplied project instructions refer to the portal course labelled “PhD with internship”; verify this course selection with the local administrator. The University is the degree institution. The University of Arizona is described as a collaborator, which does not establish a joint or dual degree. The Scottish recognised-bodies schedule lists St Andrews, consistent with the University’s own PhD programme information. Chemistry postgraduate research · Scottish recognised-bodies schedule
EastBio is a doctoral training partnership rather than the degree-awarding body. Its members include St Andrews and other East of Scotland universities and research organisations. The partnership was renewed in 2024 for five additional cohorts and offers cross-institutional expertise and training. Its current training overview describes cohort-building, research, life-sciences methods and professional skills, with core training concentrated in years one and two and research emphasis in later years. PIPS placements can give experience in a non-academic setting and new professional contacts, although the host and exact timing for this project are not stated. EastBio partnership · Training overview · Placements
St Andrews Chemistry reports that the joint EaStCHEM Chemistry submission in REF 2021 had more than 99% of outputs rated world-leading or internationally excellent, with all impact cases rated outstanding or very considerable and a 100% world-leading research-environment score. The School also reports around 160 PhD students and 100 postdoctoral researchers. These are useful indicators of a substantial disciplinary environment, though the REF submission is joint and cannot predict the experience or outcome of an individual project. QS lists St Andrews at =115 in the overall World University Rankings 2027; this is broad institutional context, not a Chemistry ranking. School of Chemistry profile · QS 2027 profile
The field is active at the interface of cyclic-dipeptide chemistry, enzyme structure and synthetic biocatalysis. A 2024 study of a different cyclodipeptide oxidase reported a filamentous architecture and argued that assembly mattered in that enzyme system. That result makes enzyme architecture a live research question but should not be transferred mechanistically to NdasCDO. Andreas and Giessen, 2024 A 2024 Chemical Society Reviews article surveys the broader role of biocatalysis in chemical synthesis. Biocatalysis review
Career evidence is broad rather than project-specific. The Royal Society of Chemistry estimates a 314,000-person UK chemical-sciences workforce and projects 6.5% sector growth over the coming decade, versus 5% for the wider labour market. Skills Development Scotland records 200 online Chemical Scientist postings from July 2024 to June 2025, down 27.9% year on year while demand was described as steady. Online postings are only a partial labour-market indicator, not a forecast of vacancies or graduate employment. These figures support the value of transferable chemistry and interdisciplinary skills but do not promise local openings. RSC workforce project · Skills Development Scotland assessment
Supervisor and Lab Assessment
Gordon Florence is a Senior Lecturer in St Andrews Chemistry. His profile describes work on rare bioactive natural products, designed analogues and multidisciplinary collaboration at the chemistry–biology interface, including biocatalytic methods. Clarissa Melo Czekster is a Reader in Biology based in the Biomedical Sciences Research Complex; her listed expertise includes enzymology, biological chemistry and structural biology. Her lab describes work in cyclic peptides, chemical biology and biocatalysis. The two supervisors therefore cover complementary parts of the advertised chemistry and enzyme research. Florence profile · Czekster profile · Czekster Lab
Three recent publications help assess fit:
| Paper | Contribution and relevance | Limit |
|---|---|---|
| Sutherland et al., 2025, Nature Communications | Directly characterizes NdasCDO and reports broad cyclic-dipeptide substrate scope. It is the most relevant publication for the advertised enzyme platform and includes both St Andrews supervisors. Paper | It does not show the proposed Barettin-inspired compounds or their 5HT2A activity. |
| Tang et al., 2025, ACS Chemical Biology | Czekster-coauthored work on biocatalysis in another enzyme and substrate family, providing evidence of recent enzyme-focused synthetic research. Paper record | It is adjacent expertise, not validation of this project’s substrate class. |
| Ogilvie and Melo Czekster, 2023, Bioorganic & Medicinal Chemistry | Review of cyclic dipeptides and the human microbiome, useful for understanding the broader chemical and biological class. Review | Review-level context, not a direct result on Barettin-derived receptor activity. |
The named University of Arizona collaborator, Christopher Cartmell, is a Research Assistant Professor of Pharmacology whose profile describes bioactive natural-product discovery. This complements the project’s pharmacology rationale, but the public description does not say whether Arizona will conduct assays, co-supervise day-to-day or host research visits. Cartmell profile
The Chemistry facilities page describes mass-spectrometry/proteomics and computational resources, and the School page says researchers can access EaStCHEM partner facilities. These pages support the presence of a broad institutional platform, not guaranteed access to a specific instrument, service, assay or collaborator laboratory. Confirm which resources are budgeted and available to this project. Facilities and services
Funding, Application, and Living Cost
EastBio’s official guidance says studentships are open worldwide and cover tuition at the home-fee level, a stipend and an annual research grant. Visa costs and the UK health surcharge are not covered. For 2026–27, UKRI publishes a minimum stipend of £21,805 and an indicative home fee level of £5,238. St Andrews lists a 2027-entry overseas postgraduate research fee of £28,140 for Chemistry within the Faculties of Science and Medicine. Its fee page says the 2027–28 home PhD fee will be set by UKRI later. Since the project page does not state whether it covers an overseas fee difference, an international applicant should obtain a written award statement before treating the place as fully funded. Do not estimate a top-up by subtracting rates for different academic years. EastBio application guidance · UKRI stipend and fee guidance · St Andrews postgraduate fees
The selection process adds a separate constraint: EastBio states that no more than 30% of studentships may go to overseas applicants. Host institutions check eligibility and completeness, project supervisors nominate candidates, and the partnership panel assesses both academic suitability for the project and fit with its interdisciplinary programme. Some supervisors may conduct an informal interview before nomination. Applicants selected for the central interview should prepare a short presentation on previous research and be ready to discuss motivation, academic potential, interdisciplinary interests, training and the professional internship. EastBio selection process
As of 6 November, the deadline of 16 November is ten calendar days away. EastBio says late applications are not accepted. The official application page requires the EastBio form, an anonymous EDI survey whose response ID is copied to the form, academic transcripts and two references on the EastBio reference form by the deadline; referees should not be project supervisors. The project-specific instructions also request English-language evidence if applicable and direct applicants to the St Andrews portal course labelled “PhD with internship”. Because the official general checklist does not explain that English document or portal label, confirm both with the local administrator. No time of day or timezone was visible in the reviewed material. How to Apply
St Andrews accommodation choices vary substantially. The current postgraduate page lists some 50-week self-catered options from £7,062, while catered and ensuite choices can be considerably more. A university cost-of-attendance table for 2026–27, published for US Federal Direct Loan administration, uses £14,313 for catered residence or rent, utilities and food, and £5,200 for personal living over 52 weeks; travel and study equipment are listed separately. These numbers are reference estimates, not a guaranteed budget or a statement of what an individual student will spend. A £21,805 UKRI minimum stipend leaves limited room for housing choice, travel and personal costs if the award does not include extra funds. Visa and health surcharge costs may also fall outside the studentship. Accommodation fees · Postgraduate research cost of attendance
Applicant Fit and Preparation
Formal requirement: EastBio states an upper-second undergraduate degree or international equivalent; a Masters degree is not required. The project description itself does not specify an undergraduate subject, English score or required prior technique. Do not assume a particular chemistry or biological qualification is a hard project-specific condition until the host confirms it.
Useful preparation: experience in organic or medicinal chemistry, biochemistry or chemical biology can help an applicant explain why the interdisciplinary project is a fit. A strong application would make the applicant’s contribution to prior research concrete, show critical reflection on what that work did and did not establish, and give evidence of learning or communicating across disciplines. These are preparation suggestions, not additional formal eligibility rules.
Interview preparation: prepare a concise research presentation, explain interest in the 5HT2A question without overstating the preclinical evidence, and show awareness of EastBio training and placement expectations. Questions about assay ownership and partner responsibilities will also test whether the proposed project has a coherent route from the enzyme work to biological evaluation. Chemistry PhD structure and normal duration are on the School’s PGR page. Chemistry postgraduate research
